abstract | This invention provides a method of isolating sex specific DNA sequences in fish that includes the steps of hybridizing an excess of denatured, sheared DNA of a first sex with a lesser amount of denatured DNA of a second sex that has been digested with a restriction endonuclease that produces sticky ends, and ligating resulting double-stranded fragments having two sticky ends derived from the DNA of the second sex into a cloning vector. This invention also provides DNA probes isolated by the preceding method. This invention also provides methods of identifying sex specific DNA sequences using the aforementioned probes or, by using PCR primers derived from the aforementioned probes. |
