titleOfInvention | Neisseria gonorrhoeae heat stable l-antigen purified of proteins and carbohydrates |
abstract | ABSTRACT OF THE DISCLOSURE Serological method for determining the presence of Neisseria gonorrhoeae antibodies in human sera and products utilized in such test. A composition containing serum is in-cubated with a composition containing a species specific antigen preparation produced from a growth culture of Neisseria gonorrhoea to form an antigen-antibody conjugate when said antibodies are present. The presence of the conjugate it then detected. The antigen is protein in nature, is stable in boiling water for a period of one hour, and is stable when incubated with the follow-ing enzymes: DNA ase, RNA ase, dextransase, neuraminidase, and lysozyme. Also the antigen has an isoelectric point of 4 + .2 and a molecular weight of active subunits of 37,000 40,000 as determined by SDS-polyacrylamide gel, contains 2-3% organic phos-phorous, and is soluble in aqueous media containing surface active agents and insoluble in methanol, chloroform, and acetone. The method of preparing the antigen preparation may include removing contaminates by precipitation of proteins with trichloroacetic acid and ultrafiltration to remove the carbohydrate. |
