abstract | PRECISION OF DISCLOSURE Establishing an early diagnosis of pregnancy in mammals in which the state of pregnancy causes excretion of chorionic gonadotropin of placental origin in the urine, is carried out using a reagent consisting of a lyophilized composition intended to be mixed with a liquid dilution immediately before use; this composition is consisting of three superposed freeze-dried layers, the layer lower comprising a substrate constituted by erythrocytes or by a synthetic polymer which can be used as substrate, on which chorionic gonadotrobin is adsorbed endowed with antigenic properties, the upper layer comprising an antiserum vis-à-vis the antigenic chorionic gonadotropin, and a middle layer interposed between the bottom layers and upper, which includes a phosphate buffer containing an agent of calcium ion chelation. We put urine in contact, possibly diluted, to be examined, with the reagent above, the reaction mixture is left to stand for? hour at 2 hours and then it is examined whether the agglutination reaction has occurred or has been inhibited. The concentration ratio in antiserum antibody / antigen is such that the reagent has a selective sensitivity threshold for gona-chorionic tropin excreted in the urine, which is greater than 1500 IU GCH / liter urine; agglutination reactions is inhibited when the urine contains more than 1500 IU of GCH / liter of urine, which is evidenced by the deposition of a ring formed by the substrate. |
