abstract | A STANDARDIZED RADIORSPIROMETRIC METHOD AND TEST KIT FOR DETECTION AND EVALUATION OF THE METABOLIC ACTIVITY OF THE SULFATE-REDUCING BACTERIA A standardized radiorespirometric method for detecting and evaluating the metabolic activity of the sulfate-reducing bacteria (BRS) which comprises, after the preparation, comprises method from BRS cultures whose metabolism is evaluated by the determination of radioactive sulfide resulting from the metabolic activity of said culture in the presence of the S ^ 35 ^ isotope, the test being carried out also in the presence of biological and radiochemical controls. The test kit for applying the method of the invention was developed in two versions, models A and model B. Model A (100), comprises a tube (10) provided with a lateral bleed tube (13) closed by a septum (11b ), which is passed through a catheter (16) to introduce solutions, nitrogen and acid. The bacterial solution (12) is placed at the bottom of the tube. The top of the tube (10) is also provided with a septum (11a). A sensor tape (14), impregnated with solutions of lead or zinc acetate, and optionally with glycerol, is fixed to the tube (10) by threads (15). Model B (200) containing a solution (40) of bacteria involves a set of two vials, one internal (20) and one external (30), which fit together. The inner flask (20) has a tapered shape, topped by a neck (22), which is closed by a first septum (23) and having in its lower portion a series of supports (24a, 24b, 24c) spaced from each other where paper filters impregnated with an absorbing solution are placed to absorb the radioactive sulfide resulting from the metabolic activity of the bacterial culture, The outer flask (30) has a substantially cylindrical shape, is equipped with a side vent (32) close to the bottom, the said lateral purge closed by a second septum (33), for the passage of a catheter (16) intended to conduct capillaries for the introduction of soups, nitrogen gas and acid, to release the radioactive hydrogen sulfide resulting from the metabolism of the BRS bacteria culture . The solution (40) containing the bacterial bacteria culture is placed in the bottom of the outer flask (30). |
