titleOfInvention method for developing polynucleotide regulatory sequences, polynucleotide regulatory sequences, use of polynucleotide regulatory sequences, expression vector, transformed or transfected cells, transfected insect larvae, method for producing recombinant proteins, use of expression vector, and use of insect larvae
abstract METHOD FOR DEVELOPING POLYNUCLEOTIC REGULATORY SEQUENCES, POLYNUCLEOTIC REGULATORY SEQUENCES, USE OF POLYNUCLEOTIC REGULATORY SEQUENCES, VECTOR OF EXPRESSION, TRANSFECTED, TRANSFECTED, TRANSFECTED, TRANSFECTED, CELLS, TRANSFECTED, CELLED The present invention provides insect-derived promoters for the expression of foreign proteins in insect cells. In the invention, polynucleotide regulatory sequences, which direct the expression of major insect proteins (hexamerins) at specific stages of larva evolution, were isolated from insects (Trichoplusia ni). These polynucleotide regulatory sequences promote the stronger expression of foreign gene in the baculovirus system when compared to that of the conventional polyhedrin promoter. In addition, the combination of the new larva-derived promoters of the invention with the pL promoter increased levels of baculovirus expression by up to 61% -375% (depending on the time of infection) compared to conventional baculoviruses used in the biotechnology industry. The pB2 promoter also directs gene expression in premature moments when compared to the polyhedrin promoter, this aspect being a great advantage for the correct protein.
