abstract | METHODS FOR SYNTHESIZING COMPOUNDS AND REPLACED MINOCYCLINE. Methods for synthesizing a carboxaldehyde-substituted minocycline compound comprising reacting a reactive minocycline intermediate under appropriate conditions with carbon monoxide, a palladium catalyst, a phosphine binder, a silane and a base, such that said tetracycline compound is replaced by carboxaldehyde is synthesized. The palladium catalyst is chosen from PdCI ~ 2 ~ (tBu ~ 2 ~ FP dichlorobis) ~ 2 ~ (di-tert-butylphenylphosphine palladium (II)] or PdCI ~ 2 ~ (DPEPhos) [palladium (II) chloride and bis (diphenylphosphinophenyl) ether]. The tetracycline reactive intermediate contains selected portions of halogens and triflates Methods of synthesizing a substituted tetracycline compound comprising reacting the carboxaldehyde-substituted tetracycline compound under palladium-catalyzed coupling conditions, hydrogenolysis conditions or conditions reductive amination. |
