abstract | RNA oligonucleotides and RNA complements replaced by hydroxymethyl The present invention is directed to RNA oligonucleotides or RNA oligonucleotide complexes, jointly referred to herein as RNA complexes, containing at least one, but optionally more than one, monomers (s) nucleotide substituted by hydroxymethyl. a hydroxymethyl-substituted nucleotide monomer means a nucleotide monomer, containing a hydroxymethyl group (which may be unsubstituted, o-substituted, for example, by a conjugation group, or converted to an optionally substituted aminomethyl group or conjugated). this hydroxymethyl group does not participate in the formation of an internucleotide bond and is not the hydroxymethyl group (containing the 5'-hydroxyl group) of a natural RNA monomer. the RNA complexes of the invention can be useful for therapeutic applications, diagnostic applications or research applications. complexes include short interfering RNA complexes (double sirens) comprising an antisense strand and a continuous or interrupted transient strand ("sense strand") capable of regulating gene expression. at least one of these filaments, possibly more than one of these filaments, are modified by one or more hydroxymethyl-substituted nucleotide monomer (s) of this invention, which can be positioned at the 3 'end, the 5' end or internally. the RNA complexes of the invention can also be single strand RNA oligonucleotides ("RNA filaments") which are modified by at least one, but optionally, more than one hydroxymethyl-substituted nucleotide monomer (s). such single-stranded strands should be considered included whenever the term complexes of strand is used in this patent application. the complexes of the invention exhibit better stability with respect to nucleolytic degradation than the corresponding complexes covered entirely from the natural monomers of RNA. |
