abstract | METHODS FOR IDENTIFYING NEW PESTICIDES GENES AND NEW GENES HOMOLOGYING WITH A TARGET GROUP OF KNOWN GENES. Methods and compositions are provided for identifying novel genes that present regions of homology with known genes of target gene groups of interest. The methods are characterized by the systematic design of oligos that are specific for regions of homology within nucleotide sequences of a known target group of genes and in performing successive rounds of PCR amplification of nucleic acid material from an organism of interest. The PCR steps are designed to identify and amplify nucleic acids containing both known and novel genes. Nucleic acid molecules containing known genes are detected and eliminated from subsequent considerations by dot blot analysis using oligonucleotide probes specific for the known genes in the target group. Potentially new genes are subjected to additional sequence analysis to confirm novelty and tested for biological activity. Compositions of the present invention include novel polynucleotides, and derived variants and fragments, consisting of novel genes and the polypeptides encoded therein. |
