abstract | <B> IMPROVED SOLUTE GLUOSE DEHYDROGENASE DEPENDENT GLUOSE DEHYDROGENASE QUINONA <D> The present invention describes a PQQ-dependent soluble glucose dehydrogenase mutant (s-GDH; EC 1.1.5.2) with improved glucose specificity compared to a malt substitution of threonine at position 348 for glycine, alanine or serine, wherein said mutant further comprises at least one mutation to enhance mutant stability and one or more mutations to enhance glucose affinity of the mutant, and / or one or more mutations for further improve the specificity of the glucose mutant compared to maltose, and wherein position 348 corresponds to the known amino acid positions of the A.calcoaceticus s-GDH wild-type sequence. Also described are genes encoding this s-GDH mutant and different applications of these s-GDH mutants, particularly to determine glucose concentration in a sample. |
