METHOD FOR PREPARING SELECTED ANTIBODIES FOR ACTIVATION OF FC RECEPTORS. The present invention relates to a method for preparing a selective antibody for the activation of the Fc region receptors (FcRs) of the antibody comprising motifs or an ITAM motif (tyrosine-based immunoreceptor activation motif), comprising the steps of obtaining monoclonal antibodies from a hybridoma, a heterohybridoma, or any human, plant or animal cell line, replacing each of His 310 and His 435 residues (Kabat numbering) in the Fc region of said antibody with a residue chosen from lysine, alanine, glycine, valine, leucine, isoleucine, proline, methionine, tryptophan, phenylalanine, serine or threonine, and then selecting antibodies by which binding to inhibitory FcRs comprising ITIM motifs (tyrosine-based immunoreceptor inhibition motive) is reduced by at least 30%, preferably by at least 50%, 70%, 80% or also by at least 90% in relation to the same anticorp which has a native Fc region. The present invention also concerns the use of an antibody, from the method of the invention, in obtaining a medicament intended in particular for the treatment of cancer and infectious pathologies. |
