abstract | ISOLATED NUCLEIC ACID MOLECULES, METHODS TO PROTECT AN INSECT PEST PLANT, METHOD FOR PROCESSING INSULATED PLANT, COMPOSITION, METHOD FOR IMPACTING AN INSECT PACKAGE Compositions and methods for protecting a plant from an insect pest are provided. Nucleic acid molecules encoding insect protoxins or insect toxins are modified to comprise at least one proteolytic activation site that is sensitive to an insect digestive protease. Cleavage of an insect protoxin, modified at the proteolytic activation site by an insect digestive protease, produces an active insect toxin in the insect pest digestive system. Cleavage of a modified insect toxin of the invention into a proteolytic activation site results in the production of an insect toxin active in the insect digestive system which exhibits increased relative pesticidal activity relative to the insect toxin not containing the insect site. proteolytic activation. Methods of using nucleic acid sequences from modified insect protoxin and modified insect toxin and the polypeptides encoded by them to protect a plant from an insect pest are provided. Particular embodiments of the invention further provide insect modified protoxin and insect toxin compositions and formulations, expression cassettes, plants, plant cells and transformed seeds. Also disclosed herein are insect digestive proteases and nucleic acid molecules encoding them. Additionally, there is further provided a method for obtaining transformed plant comprising stably transforming a plant cell with at least one polynucleotide construct, cultivating said transformed plant cell under conditions of plant growth and regenerating a transformed plant. |
