abstract Invention Patent: "METHOD FOR THE PREPARATION OF ALPHAVIRUS REPLICON PARTICLES (ARPS)". The present invention relates to methods for the production of 5 alphavirus replicon particles in high yields; replicon RNAs are subjected to electroporation in permissive cells, where the cells are in relatively high density, along with at least one helper nucleic acid that provides the necessary functions for packing. After a period of growth in the appropriate medium, the replicon particles of the alphavirus are collected from the cell surfaces in which they were produced using a salt wash in which the salt concentration is approximately 0.2 to approximately 5 M sodium chloride, calcium chloride, magnesium chloride, potassium chloride, ammonium acetate, ammonium bicarbonate, among others. After dilution, if necessary, the particles can be purified using a suitable chromatographic technique.
