The dullI (duI) corn gene of the present invention is a determinant of the endosperm starch structure. DuI mutations affect the activity of at least two enzymes involved in starch biosynthesis, namely starch synthase, SSII, and the starch branching enzyme, SBEIIa. DuI codes for a predicted protein of 1674 residues, and is expressed as a singular time pattern in the endosperm, but is not detectable in the leaf or root. The size of the DuI product and its expression pattern match precisely the known characteristics of corn SSII. The DuI product contains two different regions repeated at its unique amino terminus, one of which is identical to a conserved segment of enzymes by debranching starch. The cDNA provided by the present invention encodes SSII, and mutations within this gene affect multiple aspects of starch biogenesis by disrupting an enzyme complex containing starch synthase (s), starch branching enzyme (s), and possibly enzyme (s) ) of starch debranching. |
