abstract | it is a method for the detection of one or more polynucleotide amplification products, the method comprising, in one embodiment, the steps of: a) providing one or more oligonucleotide initiator groups, with each group comprising: i) a direct primer comprising a specific portion of fluorescence cassette and a specific portion of target 3 'downstream; and ii) one or more specific target primers; b) supply one or more extinct fluorescent pairs, each pair being characterized by: i) a first cassette oligonucleotide labeled with a chemical fluorescent portion and which has a sequence that is capable of hybridizing to the complement of the specific cassette portion of fluorescence of the direct primer of a given oligonucleotide primer group; and ii) a second cassette oligonucleotide labeled with a chemical extinction portion; c) initiate a polynucleotide isothermal amplification reaction in the presence of a strand-displacement DNA polymerase, generating a complementary sequence to at least a portion of the relevant direct primer, so that the second relevant cassette oligonucleotide is less able to hybridize with the first relevant oligonucleotide assay, by which a signal is generated; and d) detecting the signal. |
