some aspects of the present invention provide strategies, systems, reagents, methods and kits that are useful for targeted editing of nucleic acids, including editing of a single site within the genome of a cell or individual, for example, within the human genome. in some embodiments, nucleic acid programmable dna binding protein (napdnabp) fusion proteins, for example, cp1 or variants thereof, and nucleic acid publishing proteins or protein domains, for example, deaminase domains, are provided. in some embodiments, methods are provided for targeted editing of nucleic acids. in some embodiments, reagents and kits are provided for the generation of target nucleic acid publishing proteins, for example, fusion proteins from a napdnabp (eg, casx, casy, cpfl, c2c1, c2c2, c2c3 and argonaute) and proteins or nucleic acid editing domains. |
