abstract The present disclosure provides an improved method of constructing a polynucleotide library of interest in a poxvirus vector system, for example, vaccinia virus, wherein the polynucleotides of interest encode polypeptides of interest. The method comprises constructing the library in the presence of a poxvirus assembly inhibitor, for example rifampicin, which allows library construction with greater complexity and diversity than prior methods.
