abstract memantine prodrugs for transdermal sustained release, pharmaceutical compositions and uses. the present technology relates to prodrugs that allow prolonged transdermal drug delivery. prodrugs are formed by obtaining schiff bases from memantine with the carbonyl group of another molecule. the hydrolysis of these bases allows altering the drug release time. in addition to the schiff bases formed between memantine and drugs or bioactive molecules, prodrugs can comprise metal complexes that can be used as an additional modulation of the release of the active ingredient. as an embodiment of the present technology, it presents a conjugated prodrug of memantine and pyridoxal (vitamin b6) and its metal complexes of zinc(ii) and copper(ii); and conjugated prodrugs of memantine with haloperidol and other butyrophenones, such as spiperone. the technology also relates to pharmaceutical compositions containing said prodrugs and their use to prepare medicaments for the treatment of neurodegenerative diseases, such as Alzheimer's disease and schizophrenia.
