abstract | The method and kit for identifying molecular entities involved in osteoarthritis pain The present invention is in the fields of biochemistry, pharmacology, immunology, and drug development. More specifically, the invention provides an in vitro process for identifying molecular entities, such as effective therapeutic agents and / or molecular targets for drug development, for the control of chronic osteoarthritis (oa) pain. The process of the invention comprises the following steps: (i) In vitro incubation of neurons with at least one compound to stimulate inflammation, followed by (ii) analysis of a gene panel associated with pain transduction, pain transmission and / or modulation of pain responses, and (iii) correlation of the gene expression profile obtained in the previous step with oa pain and / or oa-associated inflammatory condition. In one embodiment, the process of the invention provides for in vitro interaction between chemically modified collagen and neurons derived from human cell line (s) in order to reproduce one of the early inflammatory events involved in pain genesis observed in oa. The process of the invention provides for the identification of effective therapeutic agents and / or molecular targets for drug development to control chronic pain in oa. |
