abstract | The present invention relates to the technical field of biotechnology and is characterized by the amino acid sequence qtdldylhplinswn capable of inducing the production of specific antibodies that recognize aflatoxin b1. In order to replace aflatoxin conjugates in immunizations and to propose a toxin immunization model, the mimotope-based immunogen, i.e. aflatoxin mimetic peptide expressed on the surface of bacteriophages, was obtained. The recombinant bacteriophage 3p30 was selected by the phage display technique (Fig. 1, 2 and 3) and exhibited specific binding ability to its cognate target, anti-afb1 antibody (Fig. 4), when compared to irrelevant ligands (Fig. 5). ), demonstrating dose dependent profile (Fig. 6). In immunization assay, the phage-expressed mimotope and chemically synthesized peptide were able to induce aflatoxin-recognizing anti-peptide antibodies (Fig. 7). This strategy proves the mimicry between the peptide and the toxin, allowing the application of the aflatoxin mimetic peptide for vaccine development. |
