abstract | the viability of the hydroalcoholic extract obtained from phyllanthus amarus schumn. & thonn. (eha-pa) carried in nanoemulsion as a culture medium for ovarian follicles was evaluated on the in vitro culture of porcine ovarian follicles, with diameters ranging from 150 µm to 600 µm. The nanoemulsion prepared from eha-pa was called sn-pa and was obtained with a nonionic surfactant, a vegetable oil for food use and distilled water. Follicular cultivation of the amem + treatment was performed with the follicles individually incubated in 100 µl drops under mineral oil. For the follicular cultivation of the treatment using the ne-pa bioproduct, containing the eha-pa extract, carried at low concentrations (1.0 mg to 10 mg), the follicles were incubated in 200 µl droplets in the absence of mineral oil. sn-pa and modified amen (amem +) control medium were grown for 4 days. The parameter evaluated in this experiment consisted of the follicular viability rate through vital dyes, and follicle development was evaluated by measuring its diameters. Antioxidant tests were also performed for sn-pa which used at the appropriate concentration represents an alternative medium for cell cultivation of animal ovarian follicles. |
