The present invention describes i) the construction of synthetic genes for components of a recombinant fibrin glue: Fibrinogen A, Fibrinogen B, Fibrinogen G, Factor XIIIA, prothrombin, ecarina and ERp57; ii) expression of such genes in CHO cells genetically modified to achieve high levels of expression; iii) increased fibrinogen expression by ERp57 coexpression; iv) scaling of this expression system in a bioreactor; v) purification of the glue components by chromatographic methods; vi) formulation of the glue components for application as a surgical supplement. |
