abstract | The present invention relates to the expression vector construct containing the leu2-d defective tag, which allows selection of transformant cells containing a larger number of vector copies integrated into the genome. The scope of the invention comprises an expression system containing vector set and auxotrophic yeast lineage, and the use of repeated sequences of yeast chromosome ii, chromosome iii and rdna 5s to promote homologous recombination at these loci in the yeast genome. The presence of these repeated sequences in the vector leads to their multiple integration. The auxotrophic lineage for the amino acid leucine is obtained after genetic modifications in yeast cells to disrupt the leu2 gene. The modified cells are used as hosts for the expression of genes present in vectors containing the leu2-d selection mark. The transformation of the leu2 auxotrophic lineage with the vectors of the invention enables the recovery of prototrophy when a high number of vector copies is integrated, thereby expressing the protein encoded by the gene of interest inserted into the vector. |
