The present invention relates to DNA constructs, recombinant viruses and vaccine compositions containing the obtained recombinant viruses. This invention further relates to an improvement of the attenuated live yellow fever virus expression vector 17d. The present invention provides for modifications to the heterologous protein expression cassette in the yellow fever 17d intergenic region of the vaccine virus. the two new functional domains inserted into the expression cassette were (1) an n-glycosylation motif coding sequence located between the ns1 n-terminal motif and the heterologous protein and (2) a sequence that promoted proteolytic cleavage or not. recombinant protein so as to release it from its c-terminal containing the transmembrane domains and therefore from its association with the endoplasmic reticulum membrane re. |
