titleOfInvention | mannanase production process of microbial origin using industrial by-products / waste |
abstract | Mannanase production process of microbial origin, using by-products / industrial waste mannanase are enzymes that cleave the chains of 10 mannan, present in the hemicellulosic fraction of the plant cell wall, having applications in various industries: beverages and food, paper and cellulose, textile. , soap and detergents, animal feed, among others. The present invention addresses the use of soybean hulls to obtain the mannanase enzyme by solid state (fes) and semi-solid (fess) fermentation using a non-genetically modified microorganism. The pre-inoculum of the non-genetically modified microorganism aspergillus niger lpb-28 is obtained after cultivation in 50 ml of locust bean gum agar (144 hours / 28 ° c) and inoculated (1 x 10 7 spores / g of shell). soybean) in a sterile container 20 with soybean hulls plus a nutrient solution consisting of urea (0.7 g / l), manganese sulfate (1 g / l) and sucrose (20.0 g / l), using 70 % initial moisture for the fes process (96 hours / 28 ° c), and 15% soybean hulls in the m / v proportion of nutrient solution for the fess process (144 hours / 28 ° c / under constant agitation). The best 25 mannanase yields are approximately 200 µg -1 for fes and 3600 µl -1 for fessions, reaching about 6200 µg -1 after lyophilization process. |
