titleOfInvention Plasmid psvdepolmo, cell line containing it and procedures for obtaining human recombinatory erythropoietin using the aforementioned cell line
abstract "PSVdEpoLMO PLASMID, CONTAINING CELL LINE AND PROCEDURES FOR OBTAINING HUMAN RECOMBINATORY ERYTHROPOETIN USING THE MENTIONED CELL LINE". The present invention relates to a plasmid pSVdEpoLMO that contains the Human Erythropoietin gene, with approximately twelve thousand base pairs, where the human Erythropoietin gene is under the control of the SV 40 Virus early promoter and regulatory sequences of the Molony Virus from Murine Leukemia (LTR MoMuLV), which responds to the restriction map according to Figure 1. The invention also relates to a CHOEpoEx cell line, which results from the insertion of the plasmid into CHOTK cells, which contains between 180 and 220 copies of mentioned plasmid per cell. In addition, the invention provides a procedure for obtaining human recombinatory erythropoietin, which comprises the step of selecting the producing clone by culturing the CHOEpoEx cell line in a medium containing hypoxanthine-aminopterin-thymidine (HAT), followed by the culture of selected cells in an appropriate nutrient medium.
