abstract | "HUMAN-BOVINE ATTENUATED CHIMERIC VINYL VIRUS VACCINES (PIV)". Human-bovine chimeric parainfluenza viruses (PIVs) are infectious and attenuated in humans and other mammals and are useful individually or in combination in vaccine formulations to induce an anti-PIV immune response. Isolated polynucleotide molecules and vectors incorporating a chimeric PIV genome or antigen are also provided, which includes a partial or complete human or bovine PIV antigen genome or antigenome combined or integrated with one or more of the heterologous genes or genomic segment of a different PIV. The chimeric human-bovine PIV of the invention includes an "antecedent" genome or partial or complete PIV antigenome derived from or standardized after a human or bovine PIV virus, combined with one or more heterologous genes or genomic segments of a different PIV virus to form the genome or antigenome of the chimeric human-bovine PIV. In certain aspects of the invention, the chimeric PIV incorporates an antecedent genome or partial or complete human PIV antigenome combined with one or more heterologous genes or genomic segments from a bovine PIV, according to which the resulting chimeric virus is attenuated in virtue restriction in the host range. In an alternative configuration, the chimeric human-bovine PIV incorporates an antecedent PIV genome or partial or complete bovine antigenome combined with one or more heterologous genes or genomic segment from a human PIV gene encoding a human PIV immunogenic protein, domain of protein or epitope, for example, encoded by PIV glycoprotein and / or HN genes or genomic segments. The chimeric human-bovine PIV of the invention are also useful as vectors for the development of vaccines against other pathogens. A variety of additional mutations and nucleotide modifications are provided within the chimeric human-bovine PIV of the invention resulting in a desired phenotype and structural effects. |
