abstract Disclosed is a novel and improved process for the isolationof clavulanic acid and of pharmaceutically acceptable saltsthereof such as potassium clavulanate from the aqueousfermentation broth of Streptomyced sp. P 6621 FERM P 2904, whereinfrom the fermentation broth the mycelium, a major part of proteinsand other suspended solid particles are removed by microfiltrationat a pH value of the medium between 5.8 and 6.2 and at atemperature between about 20 oC and 40 oC, the purified broth (theaqueous phase) is optionally additionally purified byultrafiltration, and the thus purified broth is concentrated byreverse osmosis, then it is directly extracted in a countercurrentin centrifugal extractors with a water-immiscrible organic solventat a pH value of the medium between 1 and 3, whereat the stillremaining proteins are simultaneously remove, from the obtainedorganic phase the water-soluble impurities are removed by washingthe organic phase with water, the organic phase is dried in vacuoin a rectification column to a water content under 0.1 vol% andthen concentrated by evaporation, decolourized by a treatment withactive carbon and then the clavulanic acid present in the dryorganic phase, most suitably in the ethyl acetate phase, isisolated and purified by a reaction withN,N'-disopropylethylenediamine at a temperature of about roomtemperature to the intermediary N,N'-disopropylethylenediammoniumdiclavulanate, which is isolated and then reacted with potassium2-ethyl hexanoate in isopropanol to yield potassium salt ofclavulanic acid having high purity.5 claims
