abstract |
Transcriptionally silent genes in a cell line or microorganisms are activated for expressionnby insertion of a DNA regulatory element that is either capable of expressing the expression of annormally expressed gene product in such cells, or is random and which binds operablynway with the quiet gene. Insertion is accomplished by homologous recombination viancreating a DNA construct comprising a target gene segment of the normal silent gene and DNAna regulatory element for gene transduction induction. This methodology is also usednto modify the expression characteristics of any endogenous gene of the givenncell line or microorganism. |
