abstract | The present patent application relates to the enzymatic synthesis of a water-insoluble glucan by a transferase reaction with a glucan-sucrase URE 13-300. The latter was obtained by culturing a recombinant strain of E. coli BL21 (DE3) containing the glucan sucrase URE 13-300 gene, which was transferred from the Leuconostoc mesenteroides strain URE 13 (registration number NBIMCC 8970, NBMPCC, Sofia). The water-insoluble glucan has a molecular weight of 2.49 x 106 Da and a radius of rotation (RG) of 60.0 to 2.78 x 106 and a radius of rotation of 67.0 and a branched structure, with a predominance of? - (1,6) linkages between glucose residues in the main chain (67.2%); 16.18%? - (1,3) connections at branch points and 8,32%? - (1,6) connections at branches. The high molecular weight glucan sucrase URE 13-300 is successfully immobilized in multilayer films with the participation of 1% chitosan and 0.2% xanthan / water-insoluble glucan, in a ratio of 1: 1 with the highest enzyme activity in a positively charged film with 12 layers and synthesizes short-chain oligosaccharides with a degree of polymerization of 4 to 6 glucose units. The synthesized glucan forms a film with antistatic properties in combination with locus bean and xanthan gum in a ratio of 1: 1: 1. |
