The invention relates to an improved method of producing recombinant blood clotting factors, in particular factor VIII and factor IX, using an immortal cell line stably expressing viral transcription activator proteins and a carrier vector having a promoter functionally linked to A DNA sequence encoding the blood clotting factor, provided that this promoter is not a viral promoter that is stimulated by these viral transcription activator proteins. The immortal human cell line carries the indicated vector and muteins of factor VIII, particularly suitable for the production method. The invention also relates to a pharmaceutical composition comprising these factor VIII muteins and to their use for the preparation of a medicament for the treatment of hemophilia. |
