The invention discloses an ARID5B gene mutation site that affects the intramuscular fat content in beef cattle, and is located at the 261bp exon 10 of the ARID5B gene. The invention employs Yanhuang cattle DNA as a template, detects the polymorphism of ARID5B gene by HRM typing technology, and then analyzes the correlation between the polymorphism of the gene and the traits for Yanhuang cattle by single-factor variance. The results of the present invention can provide a reference for improving the quality of Yanhuang cattle and screening effective genetic marker genes.
