abstract | Provided are a method for culturing primary cells of gastric cancer and gallbladder and bile duct cancer, and supporting reagents, wherein the core of the technology is: (1) treating solid tumor tissue of gastric cancer and gallbladder and bile duct cancer with a mild cell dissociation reagent, and separating primary tumor cells of gallbladder and bile duct cancer in a bile sample by means of a mild method, thereby ensuring the vitality of the cancer cells to the greatest extent; and (2) preparing a special serum-free culture medium, and performing in vitro culturing of gastric cancer and gallbladder and bile duct cancer tumor cells by means of a suspension culture system, thereby ensuring the normal expansion of cancer cells while eliminating interference from normal cells to the greatest extent. A culture of primary cells of gastric cancer and gallbladder and bile duct cancer obtained by means of the method can be used for in vitro experiments at various cell levels, second-generation sequencing, constructing an animal model, constructing a cell line, etc. It can be predicted that such a culture method has broad application prospects in the field of research and clinical diagnosis and treatment of gastric cancer and gallbladder and bile duct cancer. |
