titleOfInvention | Benzylisoquinoline alkaloids (BIA) producing microbes, and methods of making and using the same |
abstract | Aspects of the invention include host cells that are engineered to produce benzylisoquinoline nalkaloids (BlAs). The host cells include heterologous coding sequences for a variety of enzymes ninvolved in synthetic pathways from starting compounds to BIAs of the host cell. The heterologous ncoding sequences may be derived from a different source organism as compared to the host cell and nmultiple copies of the heterologous coding sequences may be present in the host cells. In some nembodiments, the host cell is selected from a reticuline-producing host cell, a sanguinarine nprecursor-producing host cell, a protoberberine-producing host cell, a thebaine-producing host cell nand an opiate-producing host cell. Also provided are methods of producing the BIAs of interest by nculturing the host cells under culture conditions that promote activity of enzymes encoded by the nheterologous coding sequences of the host cells. Aspects of the invention further include ncompositions, e.g., host cells, starting compounds and kits, etc., that find use in methods of the ninvention. |
