abstract | The present disclosure provides a primer set for amplifying multiple target DNA sequences in a sample and a method for amplifying multiple target DNA sequences in a sample with said primer set, the primer set comprises a number of pairs of upstream and downstream specific primers for each target DNA sequence, each pair of the upstream and downstream specific primers comprises a specific sequence for target sequence, and the specific sequence shall satisfy the following requirements: (1) each of the specific sequences does not amplify a sequence outside the target sequence, (2) the specific sequences do not form a dimer, and (3) the specific sequences do not form a hairpin structure, the specific sequence at its 5' terminal is linked to a universal sequence being no homologous to the genome; the specific sequence at its 3' terminal comprises on the base modification for increasing steric hindrance, and said modification does not block the binding of the specific sequence to the template perfectly matching the specific sequence and the extension, but substantially blocks the binding of the specific sequence to the template imperfectly matching the specific sequence and the extension. |
