abstract The present invention relates to a method for producing a target protein by introducing an expression vector, which contains a gene fragment containing the DNA encoding the target protein and attenuated selection marker genes and a pair of transposon sequences at both ends of this gene fragment, to buoyant mammalian cells, by inserting the gene fragment inserted between the pair of transposon sequences into the chromosome of these mammalian cells to obtain buoyant mammalian cells that produce the target protein, and by suspension culture of the mammalian cells. The present invention also relates to buoyant mammalian cells that express the target protein.
