abstract A method for quantifying nucleic acids in a sample by use of nucleic acid amplification is described, wherein before the amplification step a given amount of a known nucleic acid molecule is added as internal standard to the sample, which standard nucleic acid molecule differs from the nucleic acid to be quantified in at least one detectable feature; to achieve high accuracy and good reproducibility it is provided that, before the nucleic acid amplification, known amounts of at least two known nucleic acid molecules which differ from one another and from the nucleic acid to be quantified by at least one detectable feature are added as internal standard to the sample, the resulting amounts of amplified sample nucleic acid and standard nucleic acid are determined, and the amount of nucleic acid originally present in the sample to be quantified is determined from the resulting amounts. <IMAGE>
