abstract | Method for the identification and quantitative determination of substances which inhibit the human enzyme cyclooxygenase-2, characterized in that a) cells of a non-genetically manipulated clone of the human cell line Mono Mac 6 are stimulated with suitable growth and/or differentiation factors, the cells stimulated in this way are incubated together with potential inhibitors of cyclooxygenase-2 and after addition of arachidonic acid the concentration or amount of the products of cyclooxygenase metabolism in the culture supernatant of these cells is measured using a suitable detection method, or b) cells of a non-genetically manipulated clone of the human cell line Mono Mac 6 are incubated with potential inhibitors of cyclooxygenase-2, these cells are then stimulated with suitable growth and/or differentiation factors, and after addition of arachidonic acid the concentration or amount of the products of cyclooxygenase metabolism in the culture supernatant of these cells is measured using a suitable detection method. |
