abstract | In a process for the production of a microorganism having an insulin-encoding nucleotide sequence, in which islet cells are isolated from pancreatic tissue, mRNA is isolated from the islet cells, this mRNA is used to synthesize a cDNA having an insulin-encoding nucleotide sequence, this cDNA is reacted with a DNA transfer vector, and the reaction product of cDNA and DNA is mixed with a microorganism, the mRNA is isolated from the islet cells by homogenization of the islet cells in the presence of an RNase inhibitor preparation, thus preventing degradation of the mRNA by RNase. The cDNA is reacted with the DNA transfer vector to give an insulin- encoding nucleotide sequence. |
