titleOfInvention MEANS AND METHODS FOR CLEARING ZEARALENONE
abstract The present relates to a method of increasing the stability of an a / b-hydrolase. Furthermore, the present relates to an a / b-hydrolase obtainable by the present method. Also provided are a / b-hydrolases that have a decreased grand mean hydropathy (GRAVY) value and / or that comprise specific mutations. Furthermore, the present relates to the use of an a / b-hydrolase herein to degrade zearalenone (ZEN). Claim 1: A method for increasing the stability of an a / b-hydrolase, the a / b-hydrolase comprising a sequence corresponding to positions 145 to 218 of SEQ ID No. 1 or a sequence having 58% or more of sequence identity with a sequence corresponding to positions 145 to 218 of SEQ ID No. 1, which comprises substituting at least one amino acid in a position corresponding to a position 160 to 205 of SEQ ID No. 1 or in a position corresponding to position 159 to 204 of SEQ ID No. 2, or in a position corresponding to a position 160 to 205 of SEQ ID No. 3, 4, or 5, or in a position corresponding to a position 176 to 222 of SEQ ID No. 6, wherein the Amino acid (s) are substituted with an amino acid, which has a more negative hydropathy index than the substituted amino acid, where the hydropathy index is determined by the Kyte and Doolittle hydropathy index , thereby obtaining an a / b-hydrolase with increased stability ntada. Claim 7: An a / b-hydrolase obtainable by the method of any one of claims 4, 5 or 6. Claim 13: A composition comprising an a / b-hydrolase of any one of the preceding claims, preferably the composition is a food or feed additive or a food or feed product. Claim 15: Kit comprising the a / b-hydrolase or the composition of any one of the preceding claims.
