abstract Peptide nucleic acid derivatives targeting a pre-mRNA 3' splice site of human SNAP25 (25 kd synaptosome-associated protein) are provided. The peptide nucleic acid derivatives potently induce at least one mRNA splice variant of human SNAP25 in cells, and are useful for safely treating dermatological indications or conditions involving expression of human SNAP25 protein by topical administration. Claim 1: A peptide nucleic acid derivative represented by formula (1), or a pharmaceutically acceptable salt thereof, wherein, n is an integer between 10 and 25; the compound of formula (1) possesses at least one complementary 10-mer overlap with a 14-mer pre-mRNA sequence of [(5' ® 3') AUCCCAGGGUAACA] in the human SNAP25 pre-mRNA; the compound of formula (1) is fully complementary to human SNAP25 pre-mRNA, or partially complementary to human SNAP25 pre-mRNA with one or two mismatches; S₁, S₂, ..., Sₙ₋₁, Sₙ, T₁, T₂,n, Tₙ₋₁, and Tₙ independently represent deuterido [D], hydrido [H], substituted or unsubstituted alkyl, or substituted or unsubstituted aryl radical; X and Y independently represent hydrido radical, formyl [HC(=O)-], aminocarbonyl [NH₂-C(=O)-], aminothiocarbonyl [NH₂-C(=S)-], substituted or unsubstituted alkyl, substituted aryl substituted or unsubstituted alkylacyl, substituted or unsubstituted arylacyl, substituted or unsubstituted alkyloxycarbonyl, substituted or unsubstituted aryloxycarbonyl, substituted or unsubstituted alkylaminocarbonyl, substituted or unsubstituted arylaminocarbonyl, substituted or unsubstituted alkylaminothiocarbonyl, substituted or unsubstituted arylaminothiocarbonyl substituted, substituted or unsubstituted alkyloxythiocarbonyl, substituted or unsubstituted aryloxythiocarbonyl, substituted or unsubstituted alkylsulfonyl, substituted or unsubstituted arylsulfonyl, substituted or unsubstituted alkylphosphonyl, or substituted or unsubstituted arylphosphonyl radical; Z represents hydrido radical, hydroxy, substituted or unsubstituted alkyloxy, substituted or unsubstituted aryloxy, substituted or unsubstituted amino, substituted or unsubstituted alkyl, or substituted or unsubstituted aryl; B₁, B₂,n, Bₙ₋₁, and Bₙ are independently selected from natural nucleobases including adenine, thymine, guanine, cytosine, and uracil, and unnatural nucleobases; and, at least four of B₁, B₂,n, Bₙ₋₁, and Bₙ are independently selected from non-natural nucleobases with a substituted or unsubstituted amino radical covalently attached to the nucleobase moiety.
