ABHD11 protein has been found to exhibit diacylglycerol lipase activity provided that the N-terminal mitochondrial targeting sequence is absent. This refers therefore to a purified and enzymatically active diacylglycerol (DAG) lipase comprising a sequence at least 50% identical to SEQ ID No. 2, 3 or 4, or a sequence consisting of a fragment of at least 50 amino acids consecutive of SEQ ID No. 2, 3 or 4, wherein said DAG lipase is capable of catalyzing the hydrolysis of 1-stearoyl-2-arachidonoyl-sn-glycerol to 2-arachidonoyl glycerol. It also refers to nucleic acids encoding said DAG lipase, as well as to expression vectors and related host cells. This refers further to methods for producing said DAG lipase, to methods for screening modulators of said DAG lipase and to methods for testing the quality of a composition comprising an inhibitor of said DAG lipase. |
