| abstract | An improved system is provided for large-scale production of proteins and Ab in cell culture, in particular, in media characterized by one or more of the following characteristics: i) a cumulative concentration of amino acids greater than about 70 mM; ii) a molar ratio of accumulated glutamine to accumulated asparagine less than about 2; iii) a molar ratio of accumulated glutamine to total accumulated amino acids less than about 0.2; iv) a molar ratio of accumulated inorganic ion to total accumulated amino acids between about 0.4 and 1; or v) a combined concentration of accumulated glutamine and accumulated asparagine between approximately 16 and 36 mM. The use of such a system allows high levels of Ab production, and decreases the accumulation of certain undesirable factors, such as ammonium and lactate. Additionally, culture methods are provided that include a temperature change, which usually comprises a decrease in temperature when the culture has reached about 20% -80% of its maximum cell density. Alternatively or additionally, the present provides methods such that, after reaching a peak, the levels of lactate and ammonium in the culture decrease over time. |
