abstract A method for the mass production of human coagulation factor VII. The method comprises a) constructing an expression vector containing i) a dihydrofolate reductase promoter devoid of one of more of the CCGCC repeat sequences of the GC-rich region thereof and an operably linked dihydrofolate reductase (DHFR) gene thereto, and ii) a cytomegalovirus (CMV) promoter and a human coagulation factor VII gene operably linked thereto; b) transfect the expression vector of step a) in an animal cell line; c) culturing the transfected animal cell line from step b) in the presence of a dihydrofolate reductase inhibitor to select cells that express human coagulation factor VII with great efficiency; and d) adding sodium butyrate to the animal cells of step c), and a cell line for the mass production of human coagulation factor VII. The use of the vector containing a DHFR promoter devoid of the GC-rich repeat sequences in the method allows the human coagulation factor VII to be produced with great efficiency on a large scale, which contributes to the treatment of hemophilia. An HMF 708 cell line to produce human coagulation factor VII.
