abstract A commercial biomarker test method for the diagnosis of multiple diseases that is simple, fast, economical and promising. Hereby, biomarkers based on miRNA can be detected in human feces specimens. The method herein allows amplifying the miRNA directly from the stool specimens, without it being necessary to extract the miRNA in advance. The differential expression of a specific microRNA in the feces of patients suffering from colorectal cancer or an adenoma is indicative of the possibility of using the fecal microRNA as a potential biomarker for the detection of colorectal neoplasms. The method herein is relevant for the diagnosis, prognosis and therapy of gastroenterological / colorectal cancers and of acquired or inherited gastrointestinal diseases. Claim 1: A method for isolating and amplifying one or more microRNA (miRNA) from a biological sample, comprising the following steps: mixing a fecal sample with a solution of a salt, RNase-free water or both to form a suspension ; centrifuge the suspension; separate the supernatant from the centrifuged suspension; measure the total RNA / miRNA concentration in the supernatant using a spectrophotometer; transcribe cDNA from total RNA using one or more primers specific for miRNA; and amplifying the transcribed cDNA using a polymerase chain reaction assay (PCR), in order to obtain one or more amplified miRNAs.
