abstract | In addition a DNA construct for use in the procedure to obtain aspartic insulin. Claim 1: A method for obtaining aspartic insulin characterized in that it comprises: i) transforming a strain of Pichia pastoris yeasts with at least two expression vectors that have different selection markers from each other and wherein each vector comprises a DNA construct that encodes for an aspartic insulin precursor of formula B (1-30) -X1-Y-X2-A (1-21); ii) select among the transformed strains according to the previous stage, those recombinant strains that are MUT S and that contain more than 5 copies of the DNA construct integrated into the yeast genome; iii) select among the recombinant strains of the previous stage that which has a greater number of copies and that has a higher level of expression of the aspartic insulin precursor; iv) fermenting said recombinant strain; v) capturing the precursor of aspartic insulin from the culture medium by chromatographic techniques, vi) digesting said precursor enzymatically under suitable conditions so that at least 70% of the precursor of aspartic insulin becomes mature aspartic insulin; and vii) purify the aspartic insulin obtained from said digestion by conventional chromatographic techniques. |
