abstract The present application involves the cloning of a genetic construct comprising a sequence encoding a modified human insulin precursor under the control of a beta casein promoter in an expression vector. It also involves the transfection of the expression plasmid into bovine fetal somatic cells, such as fibroblasts, and the enucleation of bovine oocytes by nuclear transfer to generate transgenic embryos. It also leads to the generation of transgenic cattle that can produce a modified precursor of human insulin in their breast cells.
