abstract The present provides compounds and pharmaceutical compositions thereof, which are useful for modulating channel activating proteases. Such compounds are useful for treating, decreasing, or preventing a condition associated with a channel activating protease, including, but not limited to prostasin, respiratory tract diseases. Claim 1: A compound of the formula (1), or pharmaceutically acceptable salts thereof; wherein B is H, formula (2) or (CR2) k-R5; Y is -SO2-, -NHCO-, -CO- or -O-C (= O) -, with the understanding that Y is SO2 when R2 is C1-6 alkyl or phenyl; J is an optionally substituted 5 to 12 membered fused monocyclic or heterocyclic ring comprising one or more heteroatoms selected from N, O, and S; R1 is H, an optionally halogenated C1-6 alkyl, C2-6 alkenyl or C3-6 alkynyl; cyano, OH, O (CR2) lR6, SO2R6, CONR (CR2) lR6, CONR7R8 or -? - NR7R8 where R7 and R8 together with N in NR7R8 form an optionally substituted 5- to 7-membered heterocyclic ring attached to (CR2) m by means of a nitrogen atom; or R1 is an optionally substituted C3-7 cycloalkyl, aryl, or a 5- to 7-membered heterocyclic ring or hetero-aryl having no nitrogen atoms; or a residue of formula (3), wherein the P ring is an optionally substituted 5 to 7 member carbocyclic ring; R2 is C1-6 alkyl, C2-6 alkenyl, C2-6 alkynyl, aryl or wherein -L- (CR2) p-R5 wherein L is O, S, S (O), SO2 or OC (O); R3 is C1-6 alkyl, C2-6 alkenyl, C2-6 alkynyl or - (CR2) l-R5; R4 is H, C1-6 alkyl, C2-6 alkenyl, -CR = CR-R6, C2-6 alkynyl, or an optionally substituted 5-12 membered carbocyclic ring, a heterocyclic ring, aryl or hetero-aryl; or R4 a moiety of formula (4), wherein the E ring is an optionally substituted 5 to 12 membered monocyclic or fused carbocyclic or heterocyclic ring; R5 and R6 are independently an optionally substituted 5-12 membered carbocyclic ring, a heterocyclic, aryl or hetero-aryl ring; or R6 can be C1-6 alkyl or C2-6 alkenyl; each R is H, or C 1-6 alkyl, C 2-6 alkenyl, or C 2-6 alkynyl; l is from 0 to 6; and k, m, n, and p are independently from 1 to 6.
