titleOfInvention ARIL COMPOUNDS - REPLACED INDOL AND ITS METABOLITES OF THE QUINURENINE / QUINURENINE TYPE AS THERAPEUTIC AGENTS
abstract This application refers to its preparation and pharmaceutical compositions containing these compounds. The present invention also addresses the pharmaceutical use of the compounds for the inhibition of GSK3 beta kinase and the modulation of the activities of the N-methyl-D-aspartate (NMDA) channel, for the treatment of neurodegenerative disorders and other disorders. Claim 1: A compound having the formula (1) wherein: each R1, R2 and R3 are independently selected from hydrogen, carboxy, nitro, alkylsulfonyl, aminosulfonyl, aminosulfonyl alkyl, halogen, cyano, C1-4 alkyl, C1-4 alkoxy, NR'Rö, aryl, aryl-C1-4 alkyl or aryl-C1-4 alkoxy, and each of R 'and Rö is independently H or C1-4 alkyl; or R1 = Rö = CICH2CH2. or NR'Rö constitutes a saturated heterocyclic ring containing 3-8 ring members; X is: - (CH2) n-Y-, where Y is:> NH,> C = O,> C = S or none; n is 0-4; and any carbon - (CH2) n- or Y may be substituted with 1-2 substituents independently selected from halogen, carboxy, C1-4 alkyl, C1-4 alkoxy, OH, NH2 or acyl; Ar is a 3-indole, or one of its metabolites quinurenine / quinuramine where each R5 is, independently, hydrogen, halogen, C1-4 alkyl, C1-4 alkoxy, OH, NR'Rö as defined above, nitro , aryl, arylC 1-4 alkyl or arylC 1-4 alkoxy or one of its salts acceptable for pharmaceutical use; under the conditions that: if X is - (CH2) 2-NH-unsubstituted, Ar is 3- indole, R1 is 4-methylsulfonyl and R3 and R5 are hydrogen, then R2 cannot be nitro; or if X is - (CH2) 2-NH-unsubstituted, Ar is 3-indole, R1 is 4-nitro and R3 and R5 are hydrogen, then R2 cannot be 2-bromine; or if Ar is 3-indole or 2-aminobenzoyl, X is - (CH2) 2-NH- substituted or unsubstituted, R1 and R2 cannot be 2,4-dinitro; or if X is - (CH2) 2-NH- unsubstituted, Ar is 3-indo !, an R5 is 5-methoxy, then R1 and R2 cannot be 2,4-dinitro.
