abstract |
A process for preparing crystalline forms of modafinil optical enantiomers, comprising the steps consisting of: i) dissolving one of modafinil optical enantiomers in a solvent other than ethanol; ii) crystallize the modafinil enantiomer; iii) recover the crystalline form of the modafinil enantiomer thus obtained. Pharmaceutical compositions comprising the polymorphic form of one enantiomer or another; use of the polymorphic form of both enantiomers for the manufacture of a medicament for the treatment of hypersomia and central nervous system disorders. Claim 31: Method of preparing optically active modafinil from modafinic acid, characterized in that it comprises the following steps: i) to split the two optical enantiomers of the (- +) modafinic acid and recover at least one of the enantiomers; ii) contacting one of the two enantiomers obtained with a lower alkyl haloformate in the presence of alcohol and an organic base; iii) recover the product obtained; iv) convert the ester obtained to amide; v) recover the product obtained in step iv). |
