abstract | New polynucleotides isolated from prenyl protease and polypeptides encoded by polynucleotides from prenyl protease. Antibodies that are immunospecifically bound to a prenyl protease polypeptide or any derivative, variant, mutant or fragment of the prenyl potease polypeptide, polynucleotide or antibody are provided. Additionally, construction methods of transgenic plants are provided that have altered levels of polynucleotides and prenyl protease polypeptides. Methods for the identification of enzymatic substrates of prenyl protease and inhibitors are also provided. Claim 1: A method of producing a transgenic plant, which comprises introducing into a plant cell a compound that increases the activity or expression of prenyl protease to generate a transgenic cell; and regenerating a transgenic plant from said transgenic cell. Claim 2: The method of claim 1, wherein said plant has an altered phenotype selected from the group consisting of increased stress tolerance, delayed senescence, increased sensitivity to ABA, increased yield, increased productivity and increased biomass, compared to A wild type plant. Claim 11: A method of producing a transgenic plant, which comprises introducing into a plant cell a nucleic acid that inhibits the activity or expression of prenyl protease to generate a transgenic cell; and regenerating a transgenic plant from said transgenic cell. Claim 43: A method of identifying a farnesylation modulator; the method comprises: (a) providing a cell expressing the polypeptide of claim 22; (b) contacting the cell with a candidate substance; and (c) determine if the substance alters the farnesylation activity; whereby, if an alteration observed in the presence of the substance is not observed, when the cell is contacted with a composition in the absence of the substance, the substance is identified as a farnesylation modulator. |
